Abstract |
Transforming growth factor-β(TGF-β) has been considered as one of the possible gene therapy candidates for orthopedic diseases. In previous study, we demonstrated that the TGF-β gene therapy can be applicable to orthopedic patients. After transfection of TGF-β cDNA sequence to fibroblasts (NIH 3T3), stably transfected cells were injected into rabbit Achilles tendon. At 6 weeks after injection, these cells survived and stimulated the matrix formation in rabbit Achilles tendon. To evaluate the possibility of lateral epicondylitis treatment, these cells were injected into artificially made bone-tendon junction defect area. The injected cells formed a cartilage mass and compressed the tendinous portion but bony area was not stimulated. These findings suggested that the cells stimulated the surrounding cartilage forming cells but they could not stimulate osteoblasts which was surrounded by lamella bone. We think that DNA transfected cells can stimulate the surrounding cells in vivo and ex vivo gene therapy can be applicable for the regeneration of damaged tissues in orthopedic field. |
